Different families, different questions
Incretins (GIP, GLP-1) and amylin are three distinct hormone systems. Tirzepatide was engineered to act on the first two. Cagrilintide was engineered as a stabilised, long-acting form of amylin, a pancreatic peptide that is co-secreted with insulin and acts on its own receptor family. Because the two compounds touch different receptors, laboratories use them to isolate different mechanisms. A study using tirzepatide alone is an incretin study. A study using cagrilintide alone is an amylin study. A study using both is a combination-pathway study, which is the design that has driven most of the recent interest in cagrilintide.
Why cagrilintide is usually studied in combination
Most published cagrilintide work pairs it with a GLP-1-class compound rather than examining it in isolation. That is why it is often ordered alongside tirzepatide or retatrutide, and why our metabolic collection groups the three together. It does not mean cagrilintide requires a partner compound; it means the literature's centre of gravity is combination models, and a researcher planning a stand-alone amylin study will find fewer direct reference papers.
Practical differences at the bench
- Vial size and solution concentration. Cagrilintide is supplied at 5 mg per vial, tirzepatide from 15 mg. Concentration calculations after reconstitution differ accordingly; use the concentration reference.
- Half-life. Cagrilintide's reported half-life is somewhat longer, which affects the design of time-course experiments.
- Cost per milligram. Cagrilintide is the more expensive compound per milligram (about ₱470/mg at the 5 mg vial versus about ₱105/mg for tirzepatide 15 mg). Kits reduce both.
- Storage. Identical: lyophilised vials cool, dry and dark, frozen at -20°C for long storage; refrigerated at 2 to 8°C after reconstitution, no freeze-thaw cycles.
